چکیده

Abstract The molecular machinery and chromosome structures carrying out meiosis are frequently conserved from yeast to mammals. However, signals initiating appear divergent: while nutrient restriction induces in the system, retinoic acid (RA) its target Stra8 have been shown be necessary but not sufficient induce meiotic initiation mammalian germ cells. Here, we use primary culture of mouse undifferentiated spermatogonia without support gonadal somatic cells show that combination with RA is - Spo11 -dependent gene programs recapitulate transcriptomic cytologic features vivo meiosis. We demonstrate neither nor alone exerts these effects. Moreover, identify a distinctive network 11 restriction-upregulated transcription factor genes, which associated early whose expression does require RA. Our study proposes model, by upregulating key genes for program provides an vitro platform induction could facilitate research haploid gamete production.

Stra8 and its inducer, retinoic acid, regulate meiotic initiation in both spermatogenesis and oogenesis in mice.

In eukaryotes, diploid cells give rise to haploid cells via meiosis, a program of two cell divisions preceded by one round of DNA replication. Although key molecular components of the meiotic apparatus are highly conserved among eukaryotes, the mechanisms responsible for initiating the meiotic program have diverged substantially among eukaryotes. This raises a related question in animals with t...

Retinoic acid regulates sex-specific timing of meiotic initiation in mice.

In mammals, meiosis is initiated at different time points in males and females, but the mechanism underlying this difference is unknown. Female germ cells begin meiosis during embryogenesis. In males, embryonic germ cells undergo G0/G1 mitotic cell cycle arrest, and meiosis begins after birth. In mice, the Stimulated by Retinoic Acid Gene 8 (Stra8) has been found to be required for the transiti...

Methylseleninic acid synergizes with tamoxifen to induce caspase-mediated apoptosis in breast cancer cells.

Tamoxifen has efficacy as a breast cancer therapy and chemoprevention agent. However, toxicity and resistance to tamoxifen limit its clinical application. There is an urgent need to develop compounds that may be combined with tamoxifen to improve efficacy and overcome toxicity and resistance. We showed previously that the organoselenium compound methylseleninic acid (MSA) increased the growth-i...

BMP4 Cooperates with Retinoic Acid to Induce the Expression of Differentiation Markers in Cultured Mouse Spermatogonia

Spermatogenesis is sustained by the proliferation and differentiation of spermatogonial stem cells (SSCs). However, the molecules controlling these processes remain largely unknown. Here, we developed a simplified high concentration serum-containing system for the culture of mouse SSCs. Analysis of SSCs markers and transplantation results revealed that the cultured spermatogonia retained stem c...

Initiation of the Meiotic Prophase-to-metaphase Transition in Mammalian Oocytes

Mammalian oocytes are arrested in meiotic prophase for prolonged periods, and then resume meiosis in response to luteinizing hormone from the pituitary, which binds to the somatic cells surrounding the oocyte. This chapter reviews current knowledge and questions about the molecular mechanisms that mediate communication between the somatic cells and oocyte to control the prophase-to-metaphase tr...
